A Versatile Synthetic Affinity Probe Reveals Inhibitory Synapse Ultrastructure and Brain Connectivity

چکیده

Die Visualisierung von inhibitorischen Synapsen erfordert auf Sondentypen und das jeweilige bildgebende Verfahren abgestimmte Protokolle beruht in der Regel genetischer Manipulation oder Verwendung Antikörpern, die bei Gewebefärbungen unter ihrer geringen Eindringtiefe leiden. Abgestimmt Gephyrin Protein, einem universellen Marker Synapse, haben wir eine kompakte, hochaffine hochselektive peptid-basierte Sonde entwickelt. Durch Anpassung multivalenten Architektur des Farbstoffs “Sylites” erhalten; Färbungen mit hervorragendem Signal-zu-Hintergrund-Verhältnis ermöglicht, Antikörper Anwendung im Gewebe aufgrund schneller effizienter Penetration, Minderung Färbeartefakten vereinfachter Handhabung deutlich übertrifft. In hochauflösenden Mikroskopieverfahren lokalisiert Sylite präzise hemmende Synapse ermöglicht so Messungen Nanometermaßstab. Darüber hinaus zeigen wir, dass viralen Färbemethoden kombinierbar ist diese Weise nicht nur synaptische Konnektivität, sondern auch Identität Größe hemmenden Postsynapsen aufgelöst werden kann.
